Syntaxin-10 is a membrane-associated SNARE (soluble NSF attachment protein receptor) protein involved in the fusion and docking of vesicles, specifically facilitating transport from late endosomes to the trans-Golgi network[1][2][6]. It forms a SNARE complex with STX16, Vti1a, and VAMP3 that is required for proper sorting and recycling of mannose 6-phosphate receptors (MPRs), critical for lysosomal enzyme targeting in human cells[1][6]. Depletion or loss of Syntaxin-10 impairs these sorting pathways, leading to mislocalization of vesicular cargo. While highly conserved across several species, Syntaxin-10 is notably absent from mouse and rat genomes, suggesting alternate mechanisms in those species[1]. To date, there are no directly approved drugs that target Syntaxin-10, and it is primarily characterized as a vesicular transport/fusion protein rather than a receptor, enzyme, or classical therapeutic target[2][6].
