Overview

Syntaxin-12 (STX12) is a member of the syntaxin family of SNARE proteins that mediates vesicle docking and fusion within intracellular compartments, particularly the early endosomes[3][4][5]. It is a type II transmembrane protein localized mainly to endosomal membranes and is involved in the fusion and recycling of endosomes and surface proteins[4]. Together with SNARE partners such as STX6, syntaxin-12 promotes trafficking from endosomes to the plasma membrane and is essential for the organization of the endocytic pathway and recycling of cellular components[4]. SNARE proteins, including syntaxin-12, are crucial for specificity in vesicular fusion, facilitating tightly regulated exo- and endocytic processes[3]. Syntaxin-12 has predicted SNAP receptor and SNARE binding activities and is implicated in processes including autophagosome assembly, cholesterol transport, and protein recycling[5]. Although not currently a direct target of approved therapeutic drugs, disruptions in its function are relevant to diseases associated with membrane trafficking, such as neurodegeneration and infection[5][3]. *Note on aliases:* Syntaxin-13 (STX13) and syntaxin-14 (STX14) are generally considered distinct but related to syntaxin-12; occasional naming overlap may occur in older or less precise literature but should be recognized as different family members[3].
