Overview

Syntaxin 16 is a Qa-SNARE protein primarily localized to the trans-Golgi network (TGN) and is involved in vesicular trafficking and membrane fusion events, particularly mediating retrograde transport from endosomes to the TGN. It is ubiquitously expressed, but has specialized roles in certain cell types, such as regulation of E-cadherin recycling at the plasma membrane for adherens junction stability in epithelial cells, Glut4 trafficking in insulin-responsive cells, and in dendritic growth. Recent studies have also implicated syntaxin 16 in key stages of autophagy, including autophagosome formation and autolysosome biogenesis, through its interaction with other SNARE proteins and recruitment via ATG8/LC3/GABARAP family proteins. Syntaxin 16 forms complexes with other SNAREs, such as VAMP7 and SNAP-47, to mediate specific membrane fusion events essential for intracellular transport and cellular homeostasis. Mutations or depletion can disrupt cell polarity and vesicular transport but its direct linkage to disease is not yet established in the literature.
