Overview

Syntaxin 18 antisense RNA 1 (STX18-AS1) is a long non-coding RNA (lncRNA) located at chromosome 4p16, in proximity to the STX18 gene. It is not a classical therapeutic target such as a receptor, enzyme, or transporter, but instead acts as a regulator of gene expression via RNA-mediated mechanisms. Reduced STX18-AS1 expression, due to certain genetic variants, is associated with the risk of atrial septal defect (ASD), a congenital heart disease. Experimental evidence shows that STX18-AS1 has a trans-acting inhibitory effect on NKX2-5—a key cardiac transcription factor—and promotes differentiation of embryonic stem cells into cardiomyocytes. Additionally, STX18-AS1 regulates local chromatin structure and augments expression of the neighboring transcription factor gene MSX1 by antagonizing CTCF-mediated chromatin insulation. Its downregulation has been identified as a potential biomarker for ASD in fetal development, but there are currently no known drugs that directly target this lncRNA, nor are there established therapeutic mechanisms or safety concerns for such targeting[1][2][5].
