Syntaxin-binding protein 3 (STXBP3), also known as Munc18c, is a cytosolic protein that regulates membrane fusion events through interactions with syntaxin 4 and other SNARE proteins. It plays a key role in vesicle trafficking and exocytosis in multiple cell types. In endothelial and immune cells, STXBP3 participates in regulated secretion of mediators like P-selectin and von Willebrand factor, critical for vascular homeostasis and inflammation. In metabolic tissues, STXBP3 mediates insulin-stimulated vesicle trafficking and exocytosis of GLUT4-containing vesicles for glucose uptake and is necessary for the second phase of glucose-stimulated insulin secretion in pancreatic β-cells. Genetic variants and altered expression of STXBP3 are linked to diseases such as very early onset inflammatory bowel disease (VEOIBD), immune dysregulation, obesity, and sensorineural hearing loss[1][2].
