01

Overview

Syntenic Cardiovascular Conserved Region-Associated long non-coding RNA 6 (SCAR-6; F10-AS2) is a bi-exonic, polyadenylated enhancer lncRNA transcribed antisense to the 3′ UTR of the coagulation factor X (F10) gene, upstream of the PROZ gene in a highly conserved syntenic block. The locus is functionally conserved across vertebrates, with human ortholog present on chromosome 13. SCAR-6 (F10-AS2) epigenetically regulates the expression of coagulation factor genes, especially PROZ, and maintains vascular homeostasis. Loss of function (via deletion or structural alteration) leads to impaired endothelial activation, hemorrhage, and hemostatic dysfunction, highlighting its importance in cardiovascular and coagulation systems. While no approved drugs target SCAR-6 itself, it remains a significant genetic and potential therapeutic target for coagulation and vascular health, with ongoing experimental studies elucidating its complex mechanisms of action and regulatory modalities

Mechanism of action

Drugs or genetic interventions targeting SCAR-6 (F10-AS2) would act by modulating its expression or function, which in turn affects the epigenetic regulation of neighboring genes such as F10 (coagulation factor X) and PROZ (protein Z-dependent protease inhibitor)

03

Biological functions

Epigenetic regulation of gene expression (especially coagulation factors)Maintenance of vascular homeostasisModulation of hemostasisRegulation of endothelial activation through NF-κB signaling
04

Disease associations

Cardiovascular disease (e.g., vascular dysfunction, hemorrhage)Coagulation disorders (potential role due to regulation of coagulation factor genes)
05

Safety considerations

Non-coding RNAs such as SCAR-6 (F10-AS2) have pleiotropic effects, and off-target regulation could lead to unintended coagulation or vascular complicationsStructural integrity of the lncRNA is crucial (as demonstrated by functional loss after deletion), indicating challenges in designing therapeutics targeting its secondary structure
06

Interacting drugs

No approved drugs directly targeting SCAR-6 (F10-AS2) are reported; therapeutic intervention strategies for lncRNAs remain largely experimental
07

Biomarkers

Loss or mutation (e.g., the 12-bp deletion in SCAR-6) has been associated with cranial hemorrhage and vascular permeability in zebrafish, indicating possible biomarker utility for vascular stability and endothelial activation
