Overview

Synuclein alpha interacting protein (SNCAIP, commonly called Synphilin-1) is a large protein (919 amino acids) containing ankyrin-like repeats and a central coiled-coil domain, biochemical motifs that facilitate protein-protein interactions[2][1]. SNCAIP was first identified as a synaptic partner of alpha-synuclein (SNCA), with their interaction strongly implicated in the pathogenesis of neurodegenerative diseases, especially Parkinson's disease[2][1]. SNCAIP localizes primarily to synaptic terminals, participates in the formation of cytoplasmic inclusions (Lewy bodies), and modulates the ubiquitination and proteasomal degradation of alpha-synuclein, thus influencing protein aggregation and neuronal toxicity[2][1]. Several SNCAIP mutations, as well as changes in its expression and interaction with mutant alpha-synuclein variants, have been associated with altered inclusion formation, aggregation patterns, and neurodegeneration[2][1]. SNCAIP does not act as a classical enzyme, receptor, or ion channel, but rather as a protein that modulates the activity and aggregation propensity of other proteins in neurons by protein-protein interactions, particularly through its ankyrin and coiled-coil domains[2][1].
