Overview

The systemic coagulation system is a highly regulated physiological network of proteins and cellular components designed to maintain vascular integrity and prevent blood loss through the formation of fibrin clots. This system operates via a cascade of enzymatic reactions, traditionally categorized into the intrinsic and extrinsic pathways, which converge to activate thrombin and subsequently convert fibrinogen into a stable fibrin mesh (StatPearls, 2023). Beyond its primary role in hemostasis, the coagulation system is intricately linked to inflammatory processes and wound healing (PubMed, 2022). Pathological activation of this system can lead to life-threatening conditions such as deep vein thrombosis, pulmonary embolism, and ischemic stroke, while deficiencies in its components result in bleeding diatheses like hemophilia (NIH, 2021). Therapeutic intervention often involves the use of anticoagulants that target specific proteases within the cascade, such as Factor Xa or thrombin, to prevent or treat thromboembolic diseases (PubChem, 2023). Managing these therapies requires a careful balance to prevent thrombosis without inducing catastrophic bleeding complications.
