T-box transcription factor 6 (TBX6) is a phylogenetically conserved transcription factor characterized by a T-box DNA-binding domain, regulating genes involved in embryonic development. TBX6 is critical for specifying paraxial mesoderm identity and orchestrating somitogenesis, the process that produces somites—the segmented precursors of the vertebral column and ribs. Loss or mutation of TBX6 disrupts normal axial patterning and vertebral development, leading to congenital disorders such as congenital scoliosis. TBX6 functions in gene regulatory networks, interacting with other T-box family members, and represses or activates downstream targets in context-dependent manners.
