Overview

Brachyury (T-box transcription factor T) is a sequence-specific DNA-binding protein crucial for mesoderm formation and notochord development during embryogenesis. It acts as a transcriptional regulator by activating genes involved in morphogenesis and cellular differentiation. In adults, Brachyury is minimally expressed but aberrantly upregulated in several cancers, most notably chordoma. This restricted expression makes it a promising therapeutic target. Structurally, Brachyury contains a conserved T-box DNA-binding domain and dimerizes on cognate DNA sites, exerting its effects primarily by transcriptional control. Although traditionally considered “undruggable,” recent structural studies have enabled fragment-based ligand discovery efforts targeting Brachyury directly. The G177D protein variant is a biomarker for chordoma susceptibility. No approved drugs directly inhibit Brachyury, but several experimental compounds and immunotherapies are under investigation. Safety concerns center on the risk of interfering with normal developmental processes and the challenge of achieving cancer-specific inhibition.
