T cell activation by allergen extract describes the process by which CD4+ T cells recognize and respond to allergens presented by antigen-presenting cells (APCs). This interaction, mediated by the T cell receptor (TCR) and MHC class II molecules, initiates a cascade of immune events leading to either allergic inflammation or tolerance. The cytokine environment during T cell activation determines the T cell fate, driving differentiation into Th2 effector cells in allergic responses or regulatory T cells (Tregs) in tolerogenic responses. This target is central to understanding and treating allergic diseases, with immunotherapy aiming to reprogram the T cell response towards tolerance.
Allergen-specific immunotherapy aims to shift the T cell response from a Th2-dominated allergic response to a more balanced or tolerogenic response involving regulatory T cells (Tregs) and the production of blocking antibodies (e.g., IgG4). Other drugs target downstream effectors or upstream signaling pathways to reduce T cell activation or its consequences.
