A T cell activation inhibitor refers to any molecule, receptor, or pathway that suppresses the activation and proliferation of T lymphocytes. These inhibitors play a crucial role in regulating immune responses, preventing autoimmunity, and maintaining immune homeostasis by dampening excessive or inappropriate T cell activity. Key examples include CTLA-4, PD-1, LAG-3, TIM-3 and BTLA. These inhibitors can function by competing with co-stimulatory receptors, disrupting immunological synapse formation, or interfering with intracellular signaling. Clinically, checkpoint inhibitors are used in cancer immunotherapy, while other agents are used for immunosuppression in transplant rejection and autoimmune diseases. Negative regulation also stems from soluble factors (adenosine, PGE2, TGFβ) within tissue microenvironments.
