Overview

T cell activation via antigen-presenting cells (APCs) is a central process in the adaptive immune response. It involves the recognition of antigens by T cells, which are presented on the surface of APCs bound to major histocompatibility complex (MHC) molecules. This interaction triggers a cascade of intracellular signaling events that lead to T cell proliferation, differentiation, and effector function. The activation process requires two main signals: antigen recognition (TCR binding to MHC-peptide) and co-stimulation (e.g., CD28-CD80/86 interaction). Upon successful engagement, intracellular kinases are activated, gene transcription programs initiate, and cytokine production begins, driving clonal expansion and differentiation. Dysregulation can result in autoimmunity or immunodeficiency. Therapeutic modulation focuses on co-stimulatory pathways and cytokine signaling for treating autoimmune diseases and cancer.
