The T cell antigen receptor complex (TCR complex) is a multi-subunit protein assembly on T lymphocytes essential for adaptive immunity. It recognizes peptide fragments bound to MHC molecules on APCs, initiating intracellular signaling for T cell activation and immune response. The core receptor is an αβ or γδ heterodimer associated with CD3 subunits (CD3εγ, CD3εδ, CD3ζζ), crucial for signal transduction. Antigen recognition is highly specific, driven by CDR regions, particularly CDR3. Engagement triggers ITAM phosphorylation, ZAP70 recruitment, and downstream signaling. It plays a key role in detecting and eliminating infected or transformed cells. Diversity is generated via V(D)J recombination. Dysregulation can lead to immunodeficiency or autoimmunity.
