This target refers to the process by which T cells influence macrophages to adopt an M2 (alternatively activated) phenotype. M2 macrophages are associated with anti-inflammatory, tissue repair, and immunoregulatory functions, in contrast to the pro-inflammatory and microbicidal M1 phenotype. The polarization of macrophages toward the M2 state is a key mechanism in immune modulation and is relevant for therapies targeting inflammation, autoimmunity, cancer, and tissue regeneration. Key cytokines involved include IL-4, IL-13, and IL-10, acting through STAT6 and STAT3 pathways, respectively. Promoting this pathway leads to increased anti-inflammatory activity and enhanced tissue repair, while excessive activation may contribute to fibrosis or tumor progression.
