Target intelligence / Profile preview

T cell immunoreceptor with immunoglobulin and ITIM domain (TIGIT)

Target
TIGIT
Molecular classification
Immunoglobulin superfamily (IgSF), Receptor
01

Overview

TIGIT (T cell immunoreceptor with immunoglobulin and ITIM domain) is an inhibitory immune checkpoint receptor that plays a significant role in regulating immune responses, particularly in the context of cancer and autoimmunity. It is expressed on various lymphocytes and interacts with ligands like CD155 and CD112 to suppress immune cell activation and promote immune tolerance. It is a major focus for cancer immunotherapy development.

Mechanism of action

Engagement of TIGIT by its ligands leads to recruitment of phosphatases via its ITIM/ITT motifs, suppressing activation signals within T or NK cells. Competes with CD226 for ligand binding. Enhances suppressive activity of regulatory T-cells (Tregs).
