Overview

T-cell leukemia homeobox protein 3 (TLX3) is a homeobox-containing nuclear transcription factor belonging to the HOX11 subfamily. It is normally involved in embryonic brain, nervous system, and T-cell development. Deregulated expression of TLX3, often due to chromosomal translocation, is implicated in the development of pediatric T-cell acute lymphoblastic leukemia (T-ALL). In T-ALL, TLX3 acts as an oncogenic driver by promoting the proliferation and differentiation arrest of immature T-cell precursors through direct and indirect regulation of various target genes and noncoding RNAs. TLX3 may also influence neuronal subtype specification during development by acting as a context-dependent transcriptional selector gene. Its aberrant expression is considered an important molecular marker and prognostic indicator in certain leukemias
