The T cell maturation pathway is the developmental process by which hematopoietic stem cells differentiate into mature, functional T lymphocytes in the thymus. This process involves sequential rearrangement of antigen receptor genes, positive and negative selection to ensure self-tolerance, and differentiation into helper or cytotoxic T cell subsets. Disruption of this pathway can lead to immunodeficiency or autoimmunity.
Immunosuppressants interfere with T cell signaling and activation. TCR-targeting antibodies can modulate T cell activation or induce cell death. Checkpoint inhibitors enhance T cell activity by blocking inhibitory signals.
