T-cell-originated protein kinase (TOPK), also known as PBK, is a serine/threonine kinase belonging to the MAPKK family. It plays a critical role in cell division, particularly in regulating cytokinesis during mitosis. TOPK is highly expressed in various tumor tissues and is linked to enhanced proliferation, increased migration/invasion, tumor metastasis, and drug resistance. It serves as a prognostic biomarker and a therapeutic target for multiple cancers. Inhibitors like OTS514 have shown promise in suppressing cancer cell growth and enhancing sensitivity to chemotherapy.
