PTPN2 is an intracellular non-receptor type tyrosine-specific phosphatase that negatively regulates multiple signaling pathways involved in cell growth, differentiation, inflammation, and immune responses. It dephosphorylates receptor protein tyrosine kinases such as INSR, EGFR, CSF1R, and PDGFR, as well as key signaling molecules in immune cells. PTPN2 plays a significant role in immune regulation, inflammation, cancer, and potentially diabetes/autoimmunity. Loss or inhibition of PTPN2 can enhance tumor sensitivity to immune checkpoint inhibitors. Three isoforms exist due to alternative splicing.
