Overview

T-cell receptor alpha-beta expressing T cells (TCRαβ T cells) are the predominant subset of T lymphocytes in humans and most mammals, characterized by the surface expression of a heterodimeric T-cell receptor composed of an alpha (TRA) and a beta (TRB) chain, both members of the immunoglobulin superfamily. The TCRαβ complex, in association with the invariant CD3 proteins, enables T cells to recognize peptide antigens presented by major histocompatibility complex (MHC) molecules on the surface of antigen-presenting cells, initiating signal transduction pathways that result in T cell activation, proliferation, cytokine secretion, and effector differentiation. TCRαβ T cells mediate central adaptive immune responses, providing surveillance against pathogens and tumors, and modulating immune tolerance and autoimmunity. Therapies targeting TCRαβ T cells or their signaling pathways are used for immunosuppression (transplantation, autoimmune disease) and are under exploration in cancer immunotherapy and tolerance induction.
