Overview

The **T cell receptor alpha chain** (TCRα) is one of the two principal polypeptide chains (the other is the beta chain) that together form the heterodimeric T cell receptor (TCR) complex found on the surface of most T lymphocytes[2][3][5]. The TCRα chain consists of a variable (V) region—a product of V and J gene segment rearrangement—and a constant (C) region, both of which are homologous to immunoglobulin (antibody) domains[1][8]. The variable region forms a crucial part of the antigen-binding site that specifically recognizes peptide fragments presented by Major Histocompatibility Complex (MHC) molecules on antigen-presenting cells[2][3]. The diversity of the TCRα chain is generated by somatic gene recombination, allowing recognition of a vast range of antigens[8]. Together with the beta chain and associated CD3 signaling molecules, the TCRα chain relays signals that activate T cell responses, making it central to adaptive immunity and a key player in immune surveillance, infection response, cancer immunotherapy, and autoimmunity[2][6][7].
