TRAV20 encodes the variable region of the T cell receptor alpha chain, which pairs with a beta chain to form the αβ T cell receptor expressed on T lymphocytes. This receptor is responsible for recognizing processed antigenic peptides bound to major histocompatibility complex (MHC) molecules on antigen-presenting cells. The interaction mediates antigen-specific immune responses by activating T cells through signal transduction pathways involving CD3 and other adaptor proteins, ultimately resulting in T cell proliferation, cytokine release, and effector function. The variable regions (including TRAV20) contribute to the extraordinary diversity of T cell antigen specificity by undergoing genetic recombination during T cell development. TRAV20 is classified within the immunoglobulin superfamily, as part of the receptor family of molecules, and its biological functions center on antigen recognition and immune surveillance. Variability in TRAV20, as in other TCR segments, is essential for shaping the adaptive immune response.
Drugs targeting TCR generally act by modulating antigen recognition or T cell activation, including: blocking peptide-MHC engagement; inhibiting signal transduction downstream of TCR/CD3 complex; enhancing T cell activation against tumors or pathogens.
