Overview

T cell receptor alpha variable 28 (TRAV28) encodes a variable region of the TCR alpha chain, which contributes to the antigen-binding specificity of the TCR complex on T lymphocytes. The TCR is a heterodimeric membrane protein responsible for recognizing peptide antigens presented by major histocompatibility complex (MHC) molecules on antigen-presenting cells. The TCR complex, including its variable regions, is critical for triggering T cell activation, proliferation, and differentiation as part of adaptive immunity. In humans, the TRAV28 locus itself may represent a pseudogene, meaning it is present in the genome but may not encode a functional protein product; nonetheless, the broader function of the TCR alpha chain and its variable loci are essential for immune surveillance, response to infections, and are implicated in cancer immunology and autoimmunity. There are currently no drugs targeting the TRAV28 variable region specifically. In humans, the "TRAV28" gene is listed as a pseudogene on NCBI Gene, meaning it may not give rise to a functional variable region protein in the human TCR repertoire. However, it is a recognized part of the TCR alpha variable gene family, and the TCR alpha variable gene segments, as a group, are foundational in generating antigen recognition diversity for T cells.
