Overview

T cell receptor alpha variable 8-2 (TRAV8-2) refers to the variable region of the alpha chain in the T cell receptor (TCR) complex, specifically one of many gene segments that can be recombined to provide antigen specificity to individual T cells. The TCR is a membrane-bound heterodimer typically composed of alpha and beta chains, where the alpha chain contains a variable (V) and constant (C) region. TRAV8-2 is one member of the variable gene segments (Vα) contributing to the diversity of the antigen-binding site, recognizing peptides bound to major histocompatibility complex (MHC) molecules on antigen-presenting cells. The genetic rearrangement of V, J, and C segments during thymic development generates a highly diverse T cell repertoire essential for adaptive immunity. The engagement of a TCR containing TRAV8-2 with peptide/MHC leads to T cell activation through phosphorylation cascades and eventual immune responses or tolerance. Dysfunction or abnormal clonal expansion of specific TCRs, including those utilizing TRAV8-2, may be implicated in autoimmune disease, infection, and malignancy.
