Overview

T cell receptor-associated transmembrane adaptor 1 (TRAT1, also known as TRIM, TCRIM, HSPC062, pp29/30) is a transmembrane adaptor protein critical for stabilizing the T cell receptor (TCR)/CD3 complex at the surface of T cells[1][2][3][4][5][6][7]. It functions upstream of key TCR signaling events, mediates the regulation of receptor trafficking, and ensures appropriate immune cell activation. In CD8+ regulatory T cells, TRAT1 controls the localization of the CTLA-4 checkpoint protein, a central mechanism for immune suppression and tolerance. Recent research implicates TRAT1 in tumor immune microenvironments, notably in non-small cell lung cancer, where its expression may serve as a prognostic marker for cancer progression and immune therapy response. While not directly targeted by drugs, its role in immunoregulation makes it of interest for understanding and potentially manipulating immune responses in cancer and autoimmune disease[1][2][3][5][7].

Mechanism of action

For drugs that modulate T cell signaling or immune checkpoint proteins, TRAT1 contributes to stabilization of TCR complexes and presentation of regulatory molecules (such as CTLA-4), thereby affecting immune cell activation and inhibition mechanisms indirectly.
