Overview

The T cell receptor beta chain variable region family members (TCR Vβ) are gene segments encoding the variable region of the beta chain of the αβ T cell receptor. This family exists in both human and mouse genomes as a set of closely related gene segments (TRBV), each defined by specific sequence motifs and usually identified by monoclonal antibodies directed at family-specific epitopes. The TCR Vβ region determines antigen specificity by pairing with the alpha chain and forms the core antigen recognition structure that interacts with peptide-MHC complexes. Diversity in the TCR repertoire arises through random V(D)J recombination events during T-cell development, generating millions of unique antigen receptors critical for adaptive immune response. In disease contexts, specific Vβ family usage can be strongly biased according to genetic factors, disease state, antigen exposure, and clonal expansions, making these family members important both as therapeutic targets (e.g., depletion of malignant or autoreactive T cell clones) and as biomarkers for immunological diseases.

Mechanism of action

Monoclonal antibodies bind to specific TCR Vβ family epitopes on T cells, marking them for immune depletion or modulation. Chimeric immune receptors (e.g., engineered CD64 on T cells) can redirect effector cells to kill T cells expressing the targeted Vβ family with bound antibody.
