T cell receptor beta joining 2-4 (TRBJ2-4) is a gene segment that encodes the joining (J) region of the variable domain of the T cell receptor (TCR) beta chain[1][7]. The TCR beta chain is a crucial component of the αβ T cell receptor complex, which is essential for the adaptive immune response. The TRBJ2-4 segment, along with variable (V) and diversity (D) gene segments, is somatically recombined during T cell development in the thymus to generate the complementarity-determining region 3 (CDR3) of the TCR, the primary site for antigen recognition[1][3][4][5]. The resulting αβ TCR is present on T lymphocytes and is responsible for recognizing peptide-major histocompatibility (MH) complexes on antigen presenting cells, triggering T cell activation and downstream signaling pathways necessary for T cell function, growth, and differentiation[1][2][5]. TRBJ2-4 itself is not a druggable target, but rather one of many gene segments contributing to TCR diversity and function[1][5][7].
