Overview

The T-cell receptor beta variable (TCR Vβ) region is the antigen-recognizing component of the T-cell receptor complex [UniProt, 2024]. It is encoded by approximately 30 different Vβ gene families, with each T cell expressing only one family due to allelic exclusion [Maciocia et al., Nature Medicine, 2017]. In T-cell malignancies, the tumor cells are typically monoclonal and express a single Vβ family, making this epitope a highly specific therapeutic target [Karadimitris et al., 2001]. Targeting a specific Vβ family with CAR-T cells or monoclonal antibodies allows for the selective elimination of malignant T cells while sparing the majority of the healthy T-cell repertoire [Minagawa et al., 2018]. This strategy minimizes the risk of global immunosuppression and pan-T-cell aplasia seen with broader targets like CD3. Clinical applications involve identifying the patient-specific Vβ family via flow cytometry to select the appropriate targeted therapy [Beckman Coulter, 2023]. Current research is expanding the library of Vβ-specific agents to cover the most common families involved in T-cell lymphomas.
