The T cell receptor beta variable 10 (TRBV10) is a specific segment of the variable region of the T-cell receptor (TCR) beta chain. It plays a critical role in the adaptive immune system by participating in the formation of the TCR complex, which recognizes processed antigenic peptides presented by Major Histocompatibility Complex (MHC) molecules. The diversity of the TCR repertoire is generated through V(D)J recombination, and the TRBV10 family represents a specific subset of T cells within this repertoire. In clinical contexts, TRBV10 is significant because certain bacterial superantigens, such as Staphylococcal enterotoxins, specifically bind to Vβ10 regions, leading to massive, non-specific T-cell activation and potentially fatal toxic shock syndrome. Furthermore, clonal expansions of Vβ10-positive T cells are often monitored as biomarkers in autoimmune diseases and T-cell malignancies. Therapeutic strategies targeting TRBV10 include the use of monoclonal antibodies to deplete pathogenic T-cell clones or the development of TCR-based therapies for specific cancers.
