T cell receptor beta variable 10-1 (TRBV10-1) is a variable region-encoding gene segment within the human T cell receptor beta (TRB) locus, which encodes the variable domain of the beta chain in the heterodimeric T cell receptor (TCR) complex found on the surface of T cells[1][5]. The beta chain, which is assembled from distinct variable (V), diversity (D), and joining (J) gene segments through somatic V(D)J recombination, contributes to the antigen-binding specificity of TCRs by forming the complementarity-determining regions (CDRs)—mainly CDR1, CDR2 (germline-encoded), and CDR3 (recombination-generated)[1][5]. TCRs, including those containing a TRBV10-1 variable segment, allow T lymphocytes to recognize peptides presented by major histocompatibility complex (MHC) molecules, playing a central role in adaptive immunity[1][2]. The variable TRBV gene segments confer antigen diversity and specificity, but are not directly therapeutic targets themselves. Rather, detection of TRBV usage (including TRBV10-1) is used in immunology and oncology research as a biomarker for clonal T cell expansion (such as in leukemia, infection, or response to immunotherapy)[1][5]. There are no known drugs that specifically bind or modulate TRBV10-1 itself, nor are there well-established safety concerns unique to TRBV10-1. The segment’s main relevance is in its contribution to the molecular diversity and specificity of the TCR β chain[5].
