T cell receptor beta variable 10-2 (TRBV10-2) is a variable segment of the beta chain of the T cell receptor (TCR), crucial for antigen recognition and immune diversity. The TCR beta chain is encoded by multiple variable (V), diversity (D), joining (J), and constant (C) segments, and TRBV10-2 is one of the V segments selected during somatic recombination in thymic T cell development[1][4][9]. This segment contributes to the hypervariable complementarity-determining region (CDR), which directly binds antigen peptides presented by major histocompatibility complex (MHC) molecules, initiating T cell activation and adaptive immunity[1][8]. Polymorphism in TRBV genes is associated with disease susceptibility and therapeutic response, making TRBV10-2 relevant for immunological research, disease monitoring, and as a putative biomarker in immunotherapy[5][8]. However, there are no drugs currently developed to specifically target TRBV10-2, given its nature as a diversity-generating segment rather than a pharmacological receptor[5].
For drugs affecting the TCR pathway, mechanisms include antagonism or agonism of TCR-mediated signaling, T cell activation, checkpoint blockade (e.g., anti-PD-1, anti-CTLA-4). Null for direct TRBV10-2-targeting drugs.
