Overview

T cell receptor beta variable 18 (TRBV18) is a gene encoding a variable segment of the beta chain of the T cell receptor (TCR), located on human chromosome 7[2][5]. The TCR complex, including segments such as TRBV18, is essential for antigen recognition by T lymphocytes and thus mediates adaptive immune response[3][5][6]. TRBV18 is one of many Vβ gene segments recombined during T cell development to create a vast repertoire of antigen-specific TCRs. While not a druggable target itself, TRBV18 and related Vβ segments are fundamental components in the engineering of TCRs for cell-based immunotherapies (including in cancer)[1]. The relative expression or usage of TRBV18 can also be assessed as a biomarker in immune repertoire sequencing studies in several diseases, including cancer and autoimmune disorders[4]. Safety concerns chiefly relate to the specificity of engineered TCRs and the potential for cross-reactivity in therapeutic applications[1].

Mechanism of action

Antigen recognition by T cell receptor on T cells, leading to T cell activation, cytotoxicity, and immune modulation[1][5]. Therapeutic T cell engineering may exploit or modify TCRβ variable segments for tumor antigen targeting[1].

Safety considerations

Potential for off-target or cross-reactivity of engineered TCRs, leading to autoimmunity or undesired immune responses in adoptive T cell therapies[1].
