Overview

T cell receptor beta variable 21-1 is a variable region gene segment of the beta chain of the T cell receptor (TCR), a key surface protein complex on T lymphocytes responsible for the recognition of antigenic peptides presented by major histocompatibility complex (MHC) molecules[1][5]. The TCR is a heterodimer, most commonly comprised of an alpha (TRA) and a beta (TRB) chain, each with variable (V), diversity (D, beta only), and joining (J) segments assembled by V(D)J recombination, resulting in high diversity of antigen recognition[1][2]. TRBV21-1 is one of the many TRBV (T cell receptor beta variable) gene segments encoded in the TRB locus on chromosome 7q34[4]. TCR engagement is essential for T cell activation, leading to a cascade of intracellular signaling events that direct immune responses such as cytokine production and cytotoxic activity[1][5]. While TRBV21-1 itself is not directly targeted by therapeutic drugs, the TCR beta variable region contributes to antigen specificity and clonal diversity essential for adaptive immunity[2]. Abnormalities or alterations in TCR expression or repertoire, including TRBV gene usage, have been implicated in immunological diseases such as cancer, infection, and inflammation but are not commonly used as direct drug targets[1][2][5].
