T cell receptor beta variable 28 (TRBV28) is a protein coding gene encoding the variable domain of the beta chain of the T cell receptor (TCR), which is critical for antigen recognition in adaptive immunity. The TRBV28 segment forms part of the antigen binding site on αβ T cell receptors, enabling T cells to recognize peptide-MHC complexes displayed by antigen presenting cells. This recognition event triggers intracellular signaling cascades (including LCK, CD3 ITAM phosphorylation, and ZAP70 activation) that result in T cell activation, proliferation, and effector function. The Vβ domain diversity is generated by somatic recombination (V(D)J rearrangement) in the thymus, shaping the mature T cell repertoire for the immune response. TRBV28 is highly relevant in immunology, particularly in engineered receptor/cell therapies for cancer and infection, though no approved drugs are directed specifically at TRBV28. Variability in TRBV usage may have diagnostic value in monitoring T cell responses and repertoire diversity in disease contexts
