Overview

T-cell receptor beta variable 3 (TCR Vβ3), also known as TRBV28 in human IMGT nomenclature, is a specific variable segment of the T-cell receptor beta chain. It plays a critical role in the adaptive immune system by forming part of the heterodimeric TCR complex that recognizes antigenic peptides presented by MHC molecules [Source 1.1.4, 1.5.1]. In certain pathological conditions, T cells expressing the Vβ3 chain are disproportionately expanded, often representing autoreactive clones in autoimmune diseases such as rheumatoid arthritis and multiple sclerosis, or responding to superantigens like Staphylococcal enterotoxin B (SEB) [Source 1.2.1, 1.3.5]. Because of this clonal or sub-clonal expansion, TCR Vβ3 has been explored as a therapeutic target for precision immunotherapy. Strategies include TCR peptide vaccines (e.g., IR501) designed to induce tolerance, monoclonal antibodies (e.g., 5E4) to deplete specific T-cell subsets, and more recently, TCR Vβ-specific CAR-T cells for treating T-cell malignancies or severe autoimmunity [Source 1.1.1, 1.2.1, 1.4.1]. Targeting Vβ3 allows for the selective elimination of pathogenic T cells while preserving the majority of the T-cell repertoire, thereby minimizing broad immunosuppression [Source 1.1.3].
