Overview

T cell receptor beta variable 3-1 (TRBV3-1) encodes the variable domain of the beta chain of the T cell receptor (TCR), critical for adaptive immune recognition. The TCR is a membrane-bound heterodimer, usually composed of highly variable alpha and beta chains, expressed on the surface of T lymphocytes, where it specifically recognizes peptide antigens presented by the major histocompatibility complex (MHC) on antigen-presenting cells[1][2][4][5][7]. The variable region, produced by V(D)J recombination in the thymus, directly interacts with the peptide-MHC complex to mediate immune signaling, leading to T cell activation, differentiation, and regulatory immune responses[1][2][4][5][7]. The TRBV3-1 gene segment is one of many variable gene segments that contribute to the tremendous diversity of T cell receptors required for broad antigen recognition in health and disease; dysregulation or clonal expansion of T cells with certain TRBV gene usages is observed in various cancers, autoimmune diseases, and infections[1][4]. There are no approved drugs that specifically target the TRBV3-1 segment, but its variable region may serve as a biomarker in immune repertoire profiling and is of interest in T cell–based or engineered immunotherapies[1][7].
