Overview

T cell receptor beta variable 5-5 (TRBV5-5) is a protein-coded segment of the variable region of the T cell receptor (TCR) beta chain, which forms part of the antigen recognition domain of alpha-beta TCRs on T lymphocytes[1]. The TCR is integral to the adaptive immune response, allowing T cells to recognize and respond to peptide-major histocompatibility complex (pMHC) complexes presented by antigen-presenting cells[1][9]. The variable region, to which TRBV5-5 contributes, determines the specificity of antigen recognition through its highly diverse complementarity-determining regions (CDRs), generated by V(D)J recombination[1][5][9]. Genetic polymorphisms in the TRBV locus, including TRBV5-5, have been implicated in the risk of autoimmune disease and as predictive biomarkers for immune-related adverse events (irAEs) in patients receiving immune checkpoint inhibitors[2]. There are no drugs that directly target TRBV5-5, but its genetic variation affects patient stratification and management in immuno-oncology and autoimmunity[2].
