Overview

T cell receptor beta variable 6-1 (TRBV6-1) is a protein-coding gene segment encoding the variable region of the beta chain of the human T cell receptor (TCR)[4]. TRBV6-1 is one of many variable (V) gene segments that, through somatic V(D)J recombination in developing T cells, create the antigen-binding diversity necessary for recognition of peptide-major histocompatibility complex (pMHC) antigens on antigen-presenting cells[4][3]. The alpha-beta T cell receptor is a heterodimer essential for adaptive immune response, facilitating recognition of pathogenic and abnormal self antigens[3][4]. Upon binding its cognate pMHC, the TCR initiates intracellular signaling via CD3 subunits, leading to T cell activation, proliferation, and effector function[3][4]. TRBV6-1 does not represent a therapeutic target for small-molecule drugs but is a biomarker for analyzing T cell repertoire diversity and clonality in contexts such as infection, cancer, and autoimmune disease[4][3]. Changes in TRBV6-1 usage may be observed in immune monitoring studies, but this gene segment is not the target of any approved drugs or direct therapies at present[4][3].
