Overview

T cell receptor beta variable 7-4 (TRBV7-4) is a gene segment encoding the variable domain portion of the beta chain of the T cell receptor (TCR) complex, which is essential for antigen recognition by T cells and initiation of adaptive immune responses[1][3]. The TCR is a membrane-bound heterodimeric receptor—typically consisting of an alpha and a beta chain—that enables T lymphocytes to specifically recognize antigenic peptides presented by major histocompatibility complex (MHC) molecules[3]. The TRBV7-4 product forms part of the highly variable antigen-binding site, contributing to the structural diversity required for immune surveillance. Genetic diversity in TRBV segments has been implicated in individual differences in immune function and susceptibility to diseases, including autoimmunity and response to cancer immunotherapy, where certain TRBV haplotypes may predict risk of immune-related adverse events (irAEs)[4]. While no drugs target TRBV7-4 specifically, the TCR complex is a general target of immunomodulatory therapies, including checkpoint inhibitors that alter T cell activity.
