Overview

The T-cell receptor (TCR) complex is a multi-subunit integral membrane protein complex found on the surface of T lymphocytes, primarily responsible for recognizing fragments of antigen as peptides bound to major histocompatibility complex (MHC) molecules (UniProt P01848, P04234). In the context of autologous dendritic cell (DC) therapies, the TCR complex is engaged by peptide-MHC (pMHC) complexes presented on the surface of the patient's own DCs, which have been loaded with specific tumor-associated antigens (PubMed: 20671328). This engagement triggers a cascade of intracellular signaling events, including the phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) on the CD3 subunits, which ultimately leads to T-cell proliferation, cytokine secretion, and the development of cytotoxic effector functions (StatPearls: T-Cell Receptors). This mechanism is the foundation of therapeutic vaccines like Sipuleucel-T, where the goal is to prime the immune system to recognize and destroy malignant cells by specifically targeting the TCR complex with high-affinity pMHC interactions (NIH: Cancer Treatment Vaccines). While highly specific, the efficacy of targeting the TCR complex via DC engagement can be influenced by the patient's HLA type and the immunosuppressive nature of the tumor microenvironment. Safety concerns typically involve mild to moderate infusion reactions, though more potent TCR-engaging strategies can lead to systemic cytokine release or autoimmunity (PubMed: 28434943).

Mechanism of action

Activation of antigen-specific T cells through the interaction of peptide-major histocompatibility complexes (pMHC) presented on dendritic cells with the T-cell receptor (TCR) complex, leading to downstream signaling via the CD3 subunits and ZAP-70 recruitment.
