Overview

The **T-cell receptor delta chain** (TCRδ) is one of two possible variable chains (the other being alpha, α) forming the heterodimeric T-cell receptor complex found on the surface of a subset of T lymphocytes called γδ T cells. The delta chain, paired with a gamma (γ) chain, enables γδ T cells to recognize a wide variety of antigens with less dependence on classical peptide–MHC presentation than αβ T cells and is assembled in the thymus via V(D)J recombination involving variable (V), diversity (D), and joining (J) gene segments[2][3][6]. The TCRδ chain associates with the CD3 complex, and upon ligand binding, triggers T cell signaling via immunoreceptor tyrosine-based activation motifs (ITAMs), leading to cellular activation and downstream immune responses[6]. γδ T cells, via their delta chain, contribute to immune surveillance against cancers and infections, and are under active investigation for immunotherapeutic interventions[6][7]. No clinical drugs directly target only the TCRδ chain, but γδ T cells and their receptors are being developed as platforms for cell therapy in cancer and infectious diseases due to their unique tissue distribution and non-MHC–restricted antigen recognition[6].
