The Vδ2 chain is a component of the γδ T cell receptor (TCR) found predominantly in human peripheral blood. It pairs with the Vγ9 chain to form the semi-invariant Vγ9Vδ2 TCR, which is characteristic of most circulating human γδ T cells. This receptor configuration is largely unique to primates and plays a central role in immune surveillance against infections and tumors. The primary ligands recognized by this receptor are small phosphorylated metabolites called phosphoantigens (PAgs), such as HMBPP from microbes or IPP from dysregulated host metabolism. Upon activation via their TCR, these cells exhibit potent cytotoxicity and production of pro-inflammatory cytokines.
