Overview

T cell receptor delta variable 1 (TRDV1) encodes the variable (V) region of the delta chain in γδ T cell receptors, which are less common than conventional αβ T cell receptors but play important roles in mucosal immunity, cancer surveillance, and infection responses. TRDV1 is a member of the immunoglobulin superfamily and contributes to the recognition of non-peptide antigens (such as stress-induced self-antigens MICA/MICB and CD1c) at epithelial surfaces. Engagement of TRDV1-containing γδ TCRs initiates signal transduction via the CD3 complex, leading to T cell activation, cytokine release, and effector responses that are rapid and often "innate-like". Vδ1+ γδ T cells are abundant at epithelial barriers, particularly the gastrointestinal tract, and increase in frequency during certain infections and tumors, reflecting a specialized role in immune surveillance and tissue homeostasis. While no drugs target TRDV1 directly, γδ TCR-based immunotherapies are a promising area of research.
