The Vdelta2 chain is a variable region of the delta (δ) chain in the γδ T cell receptor (TCR), specifically forming part of the most common γδ TCR in human peripheral blood: the Vgamma9Vdelta2 TCR. The pairing of the Vgamma9 and Vdelta2 chains enables these cells to recognize specific non-peptide antigens, particularly phosphoantigens produced by microbes and tumor cells. Upon antigen recognition through its CDR loops—especially CDR3—the receptor triggers rapid innate-like immune responses including cytokine production (e.g., IFN-gamma, TNF-alpha). Engineering αβ T-cells to express the Vγ9Vδ2 receptor can endow them with new anti-tumor properties due to their ability to detect phosphoantigens commonly upregulated in cancerous tissues. Modified versions have been designed for dual specificity against tumors expressing both phosphoantigens and integrins such as αvβ6.
