Overview

The T cell receptor delta variable 2 chain (TRDV2) is a variable domain within the delta chain of γδ T cell receptors, especially abundant in human peripheral blood. It undergoes rearrangement during T cell development, resulting in a unique antigen recognition profile distinct from typical αβ T cell receptors. Vδ2 typically pairs with Vγ9 to form the Vγ9Vδ2 TCR complex, which recognizes non-peptide phosphoantigens and is pivotal in responses to microbial infection and tumor surveillance[2][4][6][8]. These receptors are increasingly explored as immunotherapy targets due to their capacity for rapid, broad immune activation independent of classical MHC presentation. Structural studies reveal considerable diversity and flexibility in antigen binding, which correlates with their immune sensing capabilities[4][6]. Overall, T cell receptor delta variable 2 chain is a well-defined receptor subunit, central to γδ T cell function with emerging therapeutic relevance, but not currently targeted by any approved drugs.
