Overview

T cell receptor delta variable 3 is a gene segment encoding the variable region of the δ-chain of the human γδ T cell receptor (TCR). The T cell receptor is a heterodimeric, membrane-anchored protein complex expressed on the surface of T cells; it is responsible for antigen recognition. Most T cells express either αβ or γδ TCRs, and TRDV3 forms part of the variable domain repertoire of the TCR delta chain. TRDV3 rearranges somatically with other gene segments (D and J) during T-cell development, contributing to the recognition of a diverse range of antigens, including non-peptide antigens not presented by classical MHC, such as endogenous lipids or phosphoantigens. γδ T cells, using receptors with segments like TRDV3, are positioned at epithelial boundaries, facilitate rapid immune responses, and participate in pathogen clearance and tissue repair. Their unique antigen recognition and rapid response features have led to interest in their use for immunotherapy, particularly in cancer and infection settings[1][4][5]. Note: There are currently no approved drugs that directly target TRDV3, but γδ T cell-based therapies—potentially involving TRDV3-expressing cells—are being explored in research and clinical settings[4].
