T cell receptor gamma variable 3 (TRGV3) encodes the variable domain of the γ chain of the T cell receptor (TCR), which is expressed by a subset of γδ T cells[1]. TRGV3 contributes to the antigen recognition domain in γδ T cells and is involved in binding peptide/MHC and other non-peptide antigens, enabling rapid, innate-like immune responses[1]. The γδ TCR complex containing TRGV3 is located on the external side of the plasma membrane as part of a receptor complex with CD3 subunits; upon ligand engagement, the complex triggers phosphorylation cascades that initiate T cell activation, expansion, and effector function[1][2][4]. The γδ TCRs, including those with TRGV3, display considerable conformational flexibility and are structurally and functionally distinct from the more common αβ TCRs, allowing recognition of a broader range of antigens and contributing to tissue immune surveillance[2][4]. TRGV3 itself is one of several variable gene segments generated by somatic rearrangement, contributing to the diversity of γδ T cell responses[1]. While TRGV3 and γδ T cells are being explored for roles in cancer immunotherapy and other disease settings, there are currently no drugs that selectively target TRGV3[1][4].
