T cell receptor gamma variable 5 (TRGV5) encodes the variable region of the gamma chain in the γδ T cell receptor complex, a membrane-bound heterodimer present on a subset of T lymphocytes[1][7]. The TRGV5-containing TCR is produced through somatic rearrangement of variable (V), diversity (D), and joining (J) gene segments, conferring antigen specificity[1]. TRGV5 participates in the recognition of a broad range of non-peptide antigens (including lipids and phosphoantigens), often independently of the major histocompatibility complex, and can activate rapid innate-like immune responses at epithelial barriers[1][5]. Upon antigen recognition, the TRGV5-containing TCR associates with CD3 subunits, triggering phosphorylation cascades (ITAM motifs, LCK/FYN kinases, ZAP70, LAT, PLCG1), leading to calcium mobilization and ERK activation, which in turn promote T cell expansion and differentiation[1]. Vγ5-containing TCRs can form dimeric complexes essential for potent T cell activation and may possess structural features distinct from other γδ TCRs[2][3][4]. This target has roles in immunity, infection, and cancer immunotherapy and is being studied with an eye toward immune modulation and cellular therapies[2][3].
