Overview

T cell receptor gamma variable 9 (TRGV9) encodes the variable region of the gamma chain of the human T cell receptor, which combines with delta chains (especially Vδ2) to form the Vγ9Vδ2 T cell receptor complex, a major γδ T cell subset in peripheral blood[1][2][3][5]. These receptors recognize non-peptide phosphoantigen ligands, including microbial metabolites and stress-induced self-antigens, in a manner that is major histocompatibility complex (MHC) independent[1][2]. The Vγ9Vδ2 T cells mediated by TRGV9 play critical roles in immune surveillance, rapid response to infection, tumor cell recognition, and immunoregulation[1][2][6]. The structure of Vγ9Vδ2 TCR allows significant conformational flexibility and has unique assembly characteristics within the CD3-TCR complex[1]. Alterations in Vγ9Vδ2 T cells' levels and function are associated with various diseases, including cancers, infectious diseases, and inflammatory conditions, and they are being explored as immunotherapeutic targets[6][7].
